Syllabus

Course Objectives

At the end of the course, students will be able to:Understand and describe the stages of the histological process.Apply histological and immunohistochemical staining techniques effectively.Recognize and address technical artifacts that may arise.Comprehend the fundamentals of molecular techniques as they relate to tissue samples.

Course Prerequisites

Fundamental understanding of anatomy, histology, molecular biology, and biochemistry is required. Compliance with the specified prerequisites in the study plan is mandatory.

Teaching Methods

- Lectures
- Discussion of practical cases
- Analysis of digital images and histological slides

Assessment Methods

- The oral exam is graded on a scale of thirty.
- The student must demonstrate mastery of histological, immunohistochemical, and molecular techniques, as well as the ability to critically analyze the quality of the specimens.

Texts

This is the list of recommended texts:
- "Theory and Practice of Histological Techniques" by Bancroft, 8th Edition, Elsevier (2018)
- "Basic and Advanced Laboratory Techniques in Histopathology and Cytology" by Pranab Dey, Springer (2018)
- "Anatomia Patologica, Le Basi" by Luigi Ruco and Aldo Scarpa, UTET, Scienze Mediche (2007)

Contents

Course Description
This course provides students with a solid theoretical foundation in the primary histological, immunohistochemical, and molecular techniques utilized in pathology laboratories. Special emphasis will be placed on the pre-analytical phase, including the management of biopsy and surgical specimens, the preparation and staining of histological slides, and advanced molecular biology methods applied to diagnostics and translational research.
Main Topics
Principles of optical and digital microscopy Management and acceptance of biopsy and surgical specimens Techniques for fixation, processing, paraffin embedding, and microtomy Basic histological stains (e.g., Hematoxylin-Eosin) and special stains Identification of technical artifacts and processing issues Fundamentals of immunohistochemistry: antibodies, antigen-antibody binding, and tracers Antigen retrieval and optimization techniques Immunohistochemistry staining protocols and multiplexing Automated methods and digital pathology Molecular biology techniques: FISH, CISH, DNAscope, and RNAscope.
