Syllabus

Course Objectives

The course aims to provide students with the tools to study the fundamental concepts that characterize Pharmaceutical Chemistry. Selected classes of drugs that interact with receptors, enzymes and ion channels will be examined, and the mechanisms of action and the chemical-pharmaceutical properties will be investigated. In this way the student will acquire the necessary skills to understand the problems related to the design, the mechanism of action at the molecular level, the drug-receptor interactions and the structure-activity relationships of the classes of drugs examined.

Course Prerequisites

Knowledge of the most important functional groups present in molecules of pharmaceutical interest, receptor theory, drug-receptor interaction, affinity and efficacy, agonism, competitive and non-competitive antagonism, dose-response curves, and therapeutic index. Knowledge of the relationship between physicochemical properties and biological activity, as well as geometric, optical, and conformational isomerism. Knowledge of Phase I and Phase II metabolic reactions is also expected.
The prerequisite exam for this course is Pharmaceutical and Toxicological Chemistry I.

Teaching Methods

The course consists of 48 hours of lectures, supported by slide presentations provided as teaching material on the e-learning platform. A portion of the total hours (8 hours) will be dedicated to a final review, a recap of the drug syntheses covered during the course, and a preparatory exercise for the written exam.

Assessment Methods

Student learning will be assessed through a written exam lasting one hour, consisting of 3 to 4 questions. The exam will include the description of the chemical synthesis of a drug discussed during lectures, the drawing of molecular structures related to the topics covered, with a focus on structure–activity relationships (SAR) and receptor interactions.

The assessment will also take into account the student's knowledge of Lead compound discovery and possible structural modifications aimed at Lead optimization.

The final grade will be based on the written exam and the student's ability to explain and justify any mistakes. The exam is considered passed with a minimum score of 18/30.
